For decades this was done almost entirely with formaldehyde (paraformaldehyde) fumigation, but formaldehyde is carcinogenic, corrosive, leaves a residue that needs neutralizing, and the whole process is slow and cumbersome. More labs are now switching to biosafety cabinet VHP decontamination. This guide focuses on just that — why formaldehyde is being phased out, how VHP is set up on a single cabinet, and when it’s required.

<defs><lineargradient id="cg" x1="0" y1="0" x2="0" y2="1"><stop offset="0" stop-color="#f6f9fe"/><stop offset="1" stop-color="#e6eef9"/></lineargradient>
<lineargradient id="glass" x1="0" y1="0" x2="0" y2="1"><stop offset="0" stop-color="#dcecff"/><stop offset="1" stop-color="#bcd6f5"/></lineargradient></defs>
<rect x="0" y="0" width="760" height="280" fill="url(#cg)"/>
<rect x="230" y="60" width="300" height="150" rx="10" fill="url(#glass)" stroke="#7f9dc6" stroke-width="2"/>
<rect x="240" y="70" width="280" height="60" rx="4" fill="#eef5ff" stroke="#5b7cb0"/>
<text x="380" y="105" text-anchor="middle" fill="#5a6472" font-family="Segoe UI,Arial,sans-serif" font-size="10">sealed sash opening (tape / plate)</text>
<rect x="300" y="30" width="160" height="20" rx="4" fill="#e9f0fb" stroke="#8ea6c8"/>
<text x="380" y="44" text-anchor="middle" fill="#003AA1" font-family="Segoe UI,Arial,sans-serif" font-size="10">cabinet HEPA filter</text>
<rect x="60" y="105" width="110" height="66" rx="9" fill="#002B78"/>
<text x="115" y="135" text-anchor="middle" fill="#fff" font-family="Segoe UI,Arial,sans-serif" font-size="14" font-weight="700">VHP</text>
<text x="115" y="155" text-anchor="middle" fill="#cfe0ff" font-family="Segoe UI,Arial,sans-serif" font-size="10">generator</text>

<text x="600" y="130" fill="#5a6472" font-family="Segoe UI,Arial,sans-serif" font-size="11">connected to the</text>
<text x="600" y="148" fill="#5a6472" font-family="Segoe UI,Arial,sans-serif" font-size="11">cabinet’s own ports</text>
<rect x="260" y="230" width="240" height="26" rx="5" fill="#EC001B"/>
<text x="380" y="247" text-anchor="middle" fill="#fff" font-family="Segoe UI,Arial,sans-serif" font-size="11" font-weight="700">Cabinet concentration monitored before opening</text>

Biosafety cabinet VHP decontamination: once the sash opening is sealed, a VHP generator connects to the cabinet’s own intake/exhaust ports, decontaminating the HEPA filter and interior surfaces in place.

Why formaldehyde fumigation is being phased out

Formaldehyde (usually generated by heating paraformaldehyde) was the standard for biosafety cabinet decontamination for decades, but its drawbacks are well known:

Carcinogenic and corrosive. Formaldehyde is classified as harmful to human health, carries real exposure risk for the operator, and corrodes metal and some other materials;

Residue needs neutralizing. After fumigation the residual formaldehyde has to be neutralized with ammonium bicarbonate or similar — a cumbersome step, and incomplete neutralization leaves an irritating odor;

Slow. A full formaldehyde fumigation cycle often takes hours to overnight, keeping the cabinet out of service the whole time.

VHP (vaporized hydrogen peroxide) is better on nearly every point: low temperature, the only breakdown products are water and oxygen, no neutralization step, and a cycle typically completes in anywhere from tens of minutes to two or three hours — with a validatable 6-log kill confirmed by biological indicators.
