Material can be fed directly from its transport container into the reactor, or indirectly after being weighed and sub-divided elsewhere — and in both cases the charge can arrive by gravity or by vacuum/pneumatic transfer. This guide explains the two approaches, the reactor-specific design constraints, and how to choose a containment method. It follows on from Контроль за фрезерным и микронизационным обработкой.

For potent materials, charging is usually done through a barrier isolator docked to the reactor.

Direct vs indirect charging

Direct charging transfers material straight from the transport container into the reactor. It removes intermediate steps such as central weighing and pre-dispensing — but if fine metering or weighing is needed during the charge, it can lengthen the equipment cycle time and reduce availability.

Indirect charging weighs and sub-divides the material in a dedicated weighing / dispensing room into smaller intermediate containers, which are then taken to the process area and charged. The reactor’s venting design may need careful attention to prevent a pressure rise in the receiving container.

Inert / dry atmosphere. Nitrogen is often used to keep the reactor headspace inert or dry during charging.

Charging methods

Fume hood/cabinet + drum tipper — for low-potency materials, tipping a drum or bag straight in.

Barrier isolator — for high-potency materials; may integrate a weighing scale to dispense before charging. Best suited to free-flowing materials, and often built with an integrated CIP function.

Weigh-and-charge FIBC — a weighing-equipped FIBC delivers a large batch at once (a few litres up to ~1 m³, roughly 200–400 kg per transfer), with the reactor on the floor below.

Fixed vs mobile isolator — a rail-mounted mobile isolator can serve several reactors, is removed after charging to free up access, and allows short, un-blockable transfer routes; the trade-off is the more complex closed-docking system.

Layout & GMP considerations

Space and downstream access often make it hard to couple an isolator tightly to a reactor. Cardboard or fibre drums can also raise GMP concerns in higher-grade areas. One solution is to install the isolator on the floor above and connect it by a closed pipe or chute — but long chutes can be difficult to clean or clear if they block. A mobile isolator docked to the charge port through a closed connection is another option, though it is rarely used in practice because the closed-connection system is hard to design (usually needing a lift operation) and the dispensing scale must be re-calibrated after each move.

Choosing a containment approach

The right method balances potency, batch size, ergonomics and cleaning.
