It is not enough for a cycle to <em>look<\/em> effective &mdash; efficacy has to be demonstrated with biological indicators, quantified as a log reduction, and locked into a repeatable, documented process. This guide explains how a vaporized hydrogen peroxide (VHP) cycle is validated: what the 6-log standard means, how biological indicators and the half-cycle method prove it, where indicators are placed, and how the qualified cycle is maintained over time.<\/p>\n<figure>\n<img width=\"675\" height=\"900\" decoding=\"async\" src=\"\/wp-content\/uploads\/site\/cutout-vhpgenerator.jpg\" alt=\"VHP generator used to develop and validate a repeatable bio-decontamination cycle\"><figcaption>A validated cycle depends on repeatable, measured parameters &mdash; concentration, exposure time, temperature and humidity.<\/figcaption><\/figure>\n<h2>What &#8220;validated&#8221; means for a VHP cycle<\/h2>\n<p>Validation is documented proof that a process consistently does what it is supposed to do. For VHP, that means proving the cycle reliably achieves a defined level of microbial kill on the worst-case surfaces of a specific load, using measured and repeatable parameters. It is carried out under a formal qualification framework &mdash; <strong>DQ, IQ, OQ and PQ<\/strong> (design, installation, operational and performance qualification) &mdash; with a written protocol and a signed report. The four cycle phases themselves (dehumidification, conditioning, decontamination, aeration) are described in <a href=\"\/ru\/what-is-vhp-decontamination\/\">What Is VHP Decontamination<\/a>; this article is about proving one of those cycles works.<\/p>\n<p>Crucially, efficacy is a property of <em>your<\/em> validated cycle against <em>your<\/em> load and geometry &mdash; it is established by the project&#8217;s own protocol and report, not assumed from a generic claim.<\/p>\n<h2>The 6-log standard and biological indicators<\/h2>\n<p>The accepted benchmark for sporicidal bio-decontamination is a <strong>6-log&#8321;&#8320; reduction<\/strong> &mdash; a 99.9999% reduction of a resistant challenge organism. Efficacy is demonstrated with <strong>biological indicators (BIs)<\/strong>: carriers inoculated with a known, high population of resistant bacterial spores, almost always <em>Geobacillus stearothermophilus<\/em>, at a population of 10&#8310; (one million) spores per carrier.<\/p>\n<p>Spores are used because they are the hardest form of microbial life to kill &mdash; far more resistant than the vegetative bacteria, fungi and viruses a cleanroom actually needs controlled. If a cycle reliably inactivates a million resistant spores, it comfortably covers everything less resistant. After exposure, the BIs are aseptically transferred to growth media and incubated; <strong>no growth<\/strong> confirms the spores were inactivated.<\/p>\n<h2>D-value and the half-cycle (overkill) method<\/h2>\n<p>The key quantity is the <strong>D-value<\/strong>: the exposure time needed to inactivate 90% (one log) of the spore population under set conditions. Six D-values therefore corresponds to a 6-log reduction.<\/p>\n<p>The most common approach in pharma is the <strong>half-cycle (overkill) method<\/strong>:<\/p>\n<ul>\n<li>Run cycles at <strong>half<\/strong> the intended exposure time and confirm complete kill of the 10&#8310; BIs (no growth) &mdash; typically across three consecutive successful runs.<\/li>\n<li>Complete kill of a 10&#8310; population at half-time demonstrates at least a 6-log reduction in that half.<\/li>\n<li>The <strong>routine cycle then runs at double<\/strong> that exposure &mdash; delivering an inherent safety margin (an &#8220;overkill&#8221; of &ge;12 logs) that absorbs day-to-day variation.<\/li>\n<\/ul>\n<p>This is why a validated VHP cycle is so robust: the production cycle carries roughly twice the lethality needed to pass, so normal fluctuations never compromise the result.<\/p>\n<h2>Where the indicators go: worst-case placement<\/h2>\n<p>A cycle is only validated for the locations you actually challenge, so BIs are deliberately placed at the <strong>hardest-to-reach, worst-case points<\/strong> &mdash; not the easy open surfaces. Typical positions include shadowed corners, the underside of shelves, inside mouse-holes and glove ports, behind equipment, at the far end of ducting, and within the densest part of a representative load. The <strong>load configuration is fixed and documented<\/strong>: you validate a defined loading pattern, and routine cycles must reproduce it. This is why enclosure design matters &mdash; isolators, <a href=\"\/ru\/products\/vhp-pass-box\/\">VHP pass boxes<\/a> and <a href=\"\/ru\/products\/airlock-vhp-sterilization\/\">VHP airlocks<\/a> are engineered so vapor can reach every challenged surface.<\/p>\n<h2>Chemical indicators &mdash; useful, but not proof<\/h2>\n<p>Chemical indicators (CIs) change colour when exposed to a threshold of peroxide and give a fast, visual &#8220;vapor reached here&#8221; check on each run. They are valuable for routine monitoring and troubleshooting, but a colour change only confirms <em>exposure<\/em>, not a quantified kill. <strong>CIs supplement BIs; they do not replace them.<\/strong> Only biological indicators demonstrate the actual log reduction that validation requires.<\/p>\n<h2>Keeping the cycle validated over time<\/h2>\n<p>Validation is not a one-off.
