Here is what each class means and how it maps to the Приложение 1 к GMP ЕС grades your auditors work to.

What ISO 14644-1 Defines

ISO 14644-1 classifies a cleanroom by the maximum concentration of airborne particles per cubic meter, measured at defined particle sizes (commonly ≥0.5 µm). Classes run from ISO 1 (cleanest) through ISO 9. For pharmaceutical work, the range that matters is ISO 5 to ISO 8. The lower the ISO number, the cleaner the air and the more demanding the equipment and air changes required to hold it.

ISO 14644-1 classes mapped to EU GMP grades and particle limits (at-rest).

The ISO Classes That Matter for Pharma

ISO 5 — the cleanest environment used routinely; the critical zone for aseptic filling and open product handling.

ISO 6 — sterile preparation and high-grade support.

ISO 7 — clean background/support areas surrounding the critical zone.

ISO 8 — less-critical support areas such as material airlocks and gowning.

ISO 14644-1 vs EU GMP Annex 1 Grades

EU GMP Annex 1 uses Grades A, B, C and D, and — unlike a single ISO measurement — it requires classification in two states: “at rest” and “in operation.” A room can meet a cleaner ISO class at rest but be allowed a higher particle count while people and processes are running. The at-rest mapping is shown above; in operation the limits differ:

EU GMP Grade

At rest (≈ ISO)

In operation (≈ ISO)

Grade A

ISO 5

ISO 5

Grade B

ISO 5

ISO 7

Grade C

ISO 7

ISO 8

Grade D

ISO 8

Not defined / risk-based

This “at rest vs in operation” split is why Grade A critical zones are almost always protected by a barrier system that holds ISO 5 continuously, even during intervention.

Why Cleanroom Class Drives Equipment Selection

The target class dictates the hardware. A Grade A / ISO 5 critical zone is typically maintained with an isolator or open RABS supplying unidirectional (laminar) airflow through H14 HEPA filtration. Material moving between grades passes through a VHP or dynamic pass box sized to the grade difference, and exhaust from containment areas is handled by BIBO filter housings. Get the class right first, and the equipment specification follows logically.

Building to a specific cleanroom grade?

Tell us your target ISO class / GMP grade and process, and our engineers will recommend the isolation, transfer and filtration equipment to match.

Ознакомьтесь с ассортиментом продукции
Talk to an Engineer

Cleanroom Standards ISO 14644-1 Cleanroom Classification Explained (with EU GMP Grade Comparison) By Fenggu Medical Equipment · Pharmaceutical Isolation & […]

[1

post-236","post","type-post","status-publish","format-standard","hentry","category-uncategorized
["GET
